摘要
为建立影响因素简单、接近生理状态及可重复运动的骨骼肌静力性损伤的动物模型,自制了负重站立训练实验台,选用雄性SD大鼠72只,随机分为对照组、3 d~4周实验组。将实验组大鼠前肢固定,在其后肢踝关节处施加50%体重的负荷,进行跖屈位支撑站立,每天上、下午各训练1 h,观察大鼠体重、腓肠肌常规组织学结构、超微结构以及血清CK和LDH活性的变化。结果表明,各实验组体重均显著低于对照组,腓肠肌常规组织学结构和超微结构均明显改变,且随训练时间的延长而更加显著,实验组血清CK和LDH活性亦显著高于对照组。此模型肌纤维募集方式与自然状态符合,影响因素少,运动的可重复性好。连续的静力性负荷可产生对骨骼肌微观结构的累积性损害。
To establish animal model of static skeletal muscle injury which is closer to the physiological condition and repeatable movement,the researcher invented a rat training flat of loading stand.72 male SD rats were randomly divided into control group and experimental group from 3 days to 4 weeks.It imposed the load of 50% rat's body weight on the anckle of the rats and force the hindlimb plantar flexion support stand.To observe changes of body weight,histological structure and ultrastructure of gastrocnemius,CK and LDH activity of serum,each training lasted one hour in every morning and afternoon.Results: body weight of experimental groups was significantly lower than that of control group.Histological structure and ultrastructure of gastrocnemius were significantly changed,with the extension of training time and even more significant.CK and LDH activity of serum of experimental groups was significantly higher than that of control group.Conclusion: Continuous static load through loading stand will cumulatively damage microstructure of skeletal muscle.In this model,muscle-fibrin collecting mode correspond to natural condition,less subjoin factors impact and the movement can be repeated.
