一次性离心运动及针刺对大鼠骨骼肌Wnt/β-catenin信号通路的影响 被引量:3

Effects of Eccentric Exercise and Acupuncture on the Wnt/β-catenin Signaling in Rat Skeletal Muscle

摘要 目的:通过观察离心运动及针刺干预对Wnt/β-catenin信号通路主要信号分子的影响,探讨该通路在骨骼肌损伤修复中的作用。方法:将雄性SD大鼠随机分为安静组(C)、安静针刺组(CA)、运动组(E)与运动针刺组(EA),E与EA组进行坡度为-16°的一次性跑台运动;取大鼠股中间肌,实时定量PCR检测Wnt1、Wnt3a mRNA表达,免疫印迹法检测β-catenin、GSK-3β蛋白磷酸化水平。结果:1)CA组Wnt1 mR-NA表达均低于C组,且Wnt3a mRNA在12 h、72 h及120 h显著高于C组(P<0.05);CA组72 h GSK-3β活性与120 hβ-catenin磷酸化水平显著低于C组(P<0.05)。2)EA组Wnt1、Wnt3a mRNA表达的升高程度低于E组。72 h E组和EA组GSK-3β活性与β-catenin磷酸化水平均显著低于C组(P<0.05),且E组β-catenin磷酸化水平显著低于EA组(P<0.05)。结论:一次性离心运动及运动后针刺能上调Wnt1、Wnt3a mRNA表达,降低GSK-3β活性与β-catenin磷酸化水平,针刺有上调β-catenin磷酸化水平的作用。 Objective:To observe the effects of eccentric exercise and acupuncture on Wnt/β-catenin signaling,then analyze its effects on skeletal muscle damage repairment.Methods:Male SD rats were divided into four groups:E and EA group were submitted to a single bout of eccentric exercise on a treadmill down a 16°decline.Vastus intermedius muscle was obtained in 12h,24h,48h,72h,120h post-exercise and/or after needling.Wnt1 and Wnt3a mRNA expression were detected by RT-PCR,phosphorylation of β-catenin and GSK-3β were detected by western blot.Results:1) Wnt1 mRNA expression of CA group were lower than C group,Wnt3a mRNA expression of CA group were significantly higher than C group in 12h、72h and 120h(P0.05).GSK-3β activity of CA group decreased significantly compared with C group in 72h(P0.05),phosphorylation of β-catenin also decreased significantly compared with C group in 120h(P0.05).2) Wnt1 and Wnt3a mRNA expression of EA group were lower than E group.GSK-3β activity and phosphorylation of β-catenin of E and EA group decreased significantly compared with C group in 72h(P0.05),and phosphorylation of β-catenin in E group decreased significantly than EA group(P0.05).Conclusion: A single bout of eccentric exercise and needling after exercise can up-regulate the mRNA expression of Wnt1 and Wnt3a,decrease the activity of GSK-3β and phosphorylation of β-catenin,needling has the effect on up-regulating phosphorylation of β-catenin.
