摘要
目的 :观察人工合成的 β淀粉样多肽 (Aβ)对培养神经元细胞内游离钙 ([Ca2 + ]i)的影响以及褪黑素 (Mel)的阻断作用。材料和方法 :用激光扫描共聚焦显微镜技术 ,检测给予不同浓度 (Aβ2 5~ 3 5和Mel后新生大鼠皮层培养神经元 [Ca2 + ]i荧光强度。结果 :神经元暴露Aβ2 5~ 3 50 5、 1、 10 μmol/L环境后 ,[Ca2 + ]i荧光与对照组比较均呈较有意义升高 (P <0 0 5 ) ;同时暴露Mel组 ,未见钙荧光强度升高。结论 :Aβ2 5~ 3 5可诱导神经元[Ca2 + ]i升高 ,并呈剂量依赖性 ;预先给予Mel可部分阻断这种毒性作用。
objective:To observe the effects of β-Amyloid protein(Aβ P)on the intracellular free calcium ([Ca 2+ ]i)concentration of neurons,and the blocked effect of Melatonin(Mel).Materials and Methods:[Ca 2+ ]i fluorescent intensity of culture cortical neurons derived from newborn rats were measured,after exposed Aβ 25~35 and Mel,by using Laser Scanning Confocal microscopy (LSCM) technique.Results:The [Ca 2+ ]i fluorescence intensity was increasing after expose Aβ 25~35 0 5,1 0,10 0μ mol/L( P <0 05),but while exposed Mel,observed [Ca 2+ ]i fluorescence intensity was not increasing.Conclusion:Aβ 25~35 can increase the intracellular free calcium level in cultured neurons derived from newborn rats,and were concentration dependent,Mel can block this effect.
