电针治疗对绝经后疏松症大鼠肿瘤坏死因子基因表达的影响 被引量:7

摘要 目的:从肿瘤坏死因子基因表达探讨针刺防治绝经后骨质疏松症大鼠模型的作用机制。方法:选用10~12月龄清洁级雌性SD大鼠50只。分组:假手术大鼠为对照组,10只;去势大鼠按随机数字表法分为模型组、电针组、药物(阿化磷酸钠)组、电针加药物(阿化磷酸纳)组,每组10只。假手术组不摘除卵巢,其余4组均摘除双侧卵巢,术后8周,取血液分别用放免法、电化学法、RT-PCR检测各细胞因子。结果:电针组和药物组均能不同程度降低去势SD大鼠血清TNF-α、IL-6的含量,提高血清OPG的含量,且两者功效相当。结论:电针在降低去势SD大鼠血清TNF-α、IL-6的含量,提高血清DPG的含量方面具有与药物阿仑磷酸钠相当的功效,且两者共同使用,能明显增强其功效。 Objective: To investigate the effect of electroacupuncture on the gene expression of tumor necrosis factor (TNF) in rats with menopausal osteoporosis and the prevention and treatment of this disease. Methods : Fifty clean female SD rats aged 10 - 12 months were selected as subjects. The rats receiving sham operation were assigned to control group (n = I0). Other rats were subjected to bilateral ovarian resection and were randomly divided into model group ( n = 10) , eleetroacupuneture group ( n = 10), medicine ( alendronate sodium) group ( n = 10), and electroacupuncture plus medicine group (n = 10). At 8 weeks after treatment, blood samples were collected from the rats to measure various cytokines by radioimmunoassay, electrochemical assay, and RT -PCR. Results: Both electroacupune- ture group and mediicine group showed decreased serum TNF-α and interlukin- 6 (IL- 6) levels and increased serum osteoprotegerin (OPG) level, without significant differences between the two groups. Conclusion: Electroacupuncture has equivalent efficacy to alendr- onate sodium in decreasing serum TNF - α and IL -6 levels and increasing serum OPG level in the SD rats treated by bilateral ovarian resection. Joint use of the electroacupuncture and alendronate sodium can enhance the therapeutic efficacy.
