摘要
目的 研究MxA抗病毒蛋白的表达及其活性。方法 用IFNα2b处理WI - 38细胞或人外周血单核细胞 12h或 2 4h ,并用Western -blot法或FACS法分别进行MxA蛋白表达的检测及微量细胞病变抑制法对重组的MxA和IFNα2b进行抗病毒效应实验。结果 (1)低浓度IFNα2b(>1IU/ml)可诱导WI- 38细胞表达MxA ;(2 ) 10ng/mlMxA可以抵抗 2 0TCID50 的HSV -I、Polio .V感染Vero细胞 ,抵抗 2 0 0TCID50 的VSV感染Wish细胞。结论 MxA是由I型干扰素特异诱导产生的 ,且具有广谱的直接抗病毒活性。
Objective To study the expression and antiviral activity of MxA protein. Methods WI-38 or human PBMC were induced by IFNα2b for 12 or 24 hours. The contents of MxA protein in cytoplasma of induced WI-38 or human PBMC were detected by western-blot or FACS respectively.Antiviral effect of rhMxA and IFNα2b were tested by micro cytopathogenic effect inhibition assay.Results (1)Low concentration IFNα2b(>1IU/ml) can cause expression of MxA protein in WI-38 or human PBMC.(2)10ng/ml rhMxA protein can resist 20TCID 50 HSV-I 、poliovirus in Vero cells and 200TCID 50 VSV in Wish cells.Conclusions MxA protein can be inducded exclusively by Interferon (IFNα2b) and has significant direct antiviral effect in vitro.
