摘要 目的 :探讨不同硒化合物体外抗肿瘤作用。方法 :用 MTT法测定纳米粒元素硒、低价硒和亚硒酸钠体外对人白血病细胞 HL- 60、人肺癌细胞 A549和人肝癌细胞 HC的细胞毒作用 ;用化学发光法评价这些硒化合物对体外产生活性氧的清除效果。结果 :亚硒酸钠对三种肿瘤细胞有一定细胞毒作用 ,但对体外产生的活性氧无抑制作用 ;纳米硒和低价硒 ,特别是低价硒 ,对 HL- 60细胞有强烈的杀伤作用 ,并能有效清除过氧化氢和超氧阴离子。结论 :不同硒化合物对不同肿瘤细胞有不同的细胞毒作用。纳米硒和低价硒对肿瘤细胞的杀伤作用可能与亚硒酸钠有不同的机制。 Objective:To investigate the antitumor effects in vitro of various selenium compounds such as sodium selenite,element selenium particles in nano-metre size and low valent selenium. Methods:The cytotoxicities of various selenium compounds on some tumor cells were measured using MTT assay.The scavenging actions of these selenium compounds on reactive oxygen species in vitro were valuated by chemiluminescence assay.Results:Though sodium selenite showed some cytotoxicities on HL-60,A549 and HC,which the IC 50 were respectively 2.77,1.43 and 0.98 μg selenium /ml,there were no inhibitory effect on reactive oxygen species in vitro.The cytoytoxicities of element selenium particles in nano-metre size and low-valent selenium on A549 and HC were less than that of sodium seelenate,but they showed a significant cytotoxicity on HL-60,especially low-valent selenium whose IC 50 was 0.18 μg selenium/ml,and showed a stronger suppression to hydrogen peroxide and superoxide anion radicals in vvitro.Conclusion:There were different cytotoxicty effects on different tumor cells among various selenium compounds,and perhaps there were different mechanism between element seelenium particles in nano-metre size and low-valent selenium and sodium selenite.It is indicated that the low-valent selenium had latency value applying in leukemic therapy.
