摘要
目的检测信号转导子和转录激活子1(STAT1)在鼻咽癌中的表达,探讨其在鼻咽癌发生过程中的作用及其临床意义。方法采用SYBR GreenI荧光定量RT-PCR技术对48例鼻咽癌组织和12例慢性鼻咽炎组织中STAT1 mRNA的表达进行检测,并分析其表达情况与临床病理资料的相关性。结果鼻咽癌癌组织中STAT1 mRNA的表达水平明显高于慢性鼻咽炎组织(P<0.05);在肿瘤TNM分期中,T3+T4期癌组织STAT1 mRNA表达水平明显高于T1+T2期(P<0.05);在临床分期中,Ⅲ+Ⅳ期癌组织STAT1 mRNA表达水平明显高于Ⅰ+Ⅱ期(P<0.05);而其表达与有无淋巴结转移无明显相关(P>0.05)。结论 STAT1 mRNA在鼻咽癌组织中高表达,其表达增强可能与鼻咽癌的发生和发展相关,可作为鼻咽癌恶性潜能的检测指标之一。
Objective To investigate the expressionof STAT1 and itscorrelationwiththe clinical pathological parameters in nasopharyngeal carcinoma,and to reveal the potential mechanism of STAT1 underlying nasopharyngeal carcinoma.Methods The technique of quantitative reverse transcription polymerasechain reaction was applied to detect the STAT1 mRNA expression in 48 cases of nasopharyngeal carcinomatissues and 12 cases of normal nasopharyngeal mucosa.mRNA expression level of STAT1 in different groups was compared statistically.Results The expression of STAT1 mRNA was significantly higher in nasopharyngeal carcinoma tissues than that in normal nasopharyngeal mucosa(P 0.05),Besides,the relative mRNA expression of STAT1 was significantly higher in stage T3 + T4 cancer group than that in stage T1 + T2 group(P 0.05),and was significantly higher in clinical stage Ⅲ + Ⅳ cancer group than that in stage Ⅰ + Ⅱ group(P 0.05),but no association with lymphocyte metastasis(P 0.05).Conclusion STAT1 mRNA expression up-regulated in nasopharyngeal carcinoma might relates to tumor occurrence and development,which may serve as a biomarker for invasiveness of nasopharyngeal carcinoma.
