脂肪间充质干细胞对辐射损伤大鼠丙二醛及抗氧化酶的影响 被引量:1

Effects of Adipose-derived Stem Cells on Antioxidant Enzymes and Malondialdehyde in Radiation Damaged Rat

摘要 通过建立60Coγ射线照射诱发辐射损伤动物模型,移植脂肪间充质干细胞(ADSCs)后,在实验不同阶段检测SD大鼠血清抗氧化酶活力和脂质过氧化物水平。结果表明,第4天和第11天,单纯照射组丙二醛含量升高显著,是ADSCs植入组的2.00和1.58倍;抗氧化酶方面,ADSCs植入组的超氧化物歧化酶和谷胱甘肽过氧化物酶活力均高于单纯照射组,第11天差异显著。结果提示,60Coγ射线照射可导致生物体脂质过氧化物水平增高,ADSCs的移植促进机体抗氧化防御系统清除活性氧自由基的能力,恢复活性氧自由基与抗氧化防御系统之间的动态平衡,说明ADSCs在辐射损伤的治疗修复中起到积极有效作用。 Through the establishment of the damaged animal model induced by 60Co gamma-ray irradiation, malondialdehyde (MDA) content and antioxidant enzymes activity in the serum in rats were determined at three stages (the 4-th day, the ll-th day and the 33-th day after adipose-derived stem cells (ADSCs) transplantation). The results showed that MDA levels at the radiation group increased 1.29, 2.02 times the control group and 2.00, 1.58 times the ADSCS transplantation group, respectively. There was significant difference between the ADSCs transplantation group and the radiation group. For antioxidant enzymes, Superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity of the radiation group was lower than that of control group. There were significant differences between them at the ll-th day. Howerer, SOD and GSH-Px activity of ADSCs transplantation group was higher than that of the radiation group, and at the 11-th day there was also significant difference between them. At the 33th day, no significant differences were observed at all tested groups at the levels of MDA, SOD and GPx. The conclusion showed that 60Co gamma-ray radiation damage re- suhed in an increase at lipid peroxide content. And the results also indicated that ADSCs transplantation can promote the ability to eliminate reactive oxygen free radicals system (ROS), therefore, to restore the dynamic balance of ROS and antioxidant defense system. The present study indicated that ADSCs might play a positive role in radiation damage recovery and radiation therapy.
