琼脂糖电泳中核酸染料Goldview最佳使用方案的建立 被引量:3

摘要 目的确定琼脂糖凝胶电泳中使用核酸染料Goldview的最佳浓度和方法,尽可能减少实验中Goldview用量,控制其对实验室环境的污染。方法配置含不同浓度Goldview的上样缓冲液,混合不同浓度DNA溶液的琼脂糖凝胶电泳,然后对比预染样品法、前染法及后染法的染色效果。结果实验结果显示上样缓冲液中含0.4%Goldview的预染样品法电泳检测核酸的效果最佳且能检测的核酸样品浓度在16.5ng或以上为最佳。结论核酸染料Goldview在核酸的琼脂糖凝胶电泳中使用的最佳方案为预染样品法,实验中推荐浓度达到普通实验要求,并且最易控制对周围环境的污染。 Objective To confirm the optimal concentration and methods of nucleic acid dyes Goldview used in the agarose gel electrophoresis to reduce the amount of Goldview and further control its pollution to the laboratory environment. Methods Different concentrations of Goldview in the loading buffer were prepared and mixed with different concentrations of DNA solution in the agarose gel electrophoresis. The pre- staining, routine nucleic acid staining and staining after electrophoresis were compared. Results The loading buffer containing 0.4% Goldview in the pre - staining method was best, in which the opti- mal concentration in the nucleic acid samples was 16.5 ng or above. Conclusion The optimal scheme of the nucleic acid dyes Goldview in agarose gel electrophoresis is the pre - staining method with the rec- ommended concentrations to meet the common experiment requirement and the easiest way to control the pollution to the surrounding environment.
