用地高辛标记工程菌DNA片断作探针 ,通过分子杂交和免疫显色检测重组天冬酰胺酶半成品中的残余DNA。该法对半成品用氯仿抽提 ,消除了蛋白质对DNA测定的干扰。 3批重组门冬酰胺酶半成品的测定结果表明 ,其残余DNA均小于 10 0 pg/剂量。

Residual cell DNA in the products of recombinant L-Asparaginase Ⅱ was detected by hybridization with digoxigenin labeled total DNA of engineering bacteria E coli The samples were extracted by chloroform in order to remove the protein which interfere the determination of residue cell DNA The results showed that the content of DNA in three batches are all lower than 100 pg/dose.
