闽南乌龙茶茶树种质资源RAPD指纹图谱构建及遗传多样性分析 被引量:8

摘要 利用RAPD分子标记技术构建了30份闽南乌龙茶茶树种质的DNA指纹图谱,结果表明使用同一扩增引物的特异谱带类型和不同扩增引物的谱带类型组合可以有效鉴别闽南乌龙茶茶树种质资源;10条RAPD引物共扩增出103条谱带,其中80条具有多态性,占77.67%。通过UPGMA法聚类分析,30份茶树种质被聚为2个类群,供试种质间的平均遗传相似系数为0.603,遗传关系树状图在分子水平上清楚的显示了闽南乌龙茶茶树种质资源间的亲缘关系,为评估供试样品的遗传演化地位并从中选择适宜种质作为亲本进行茶树育种研究提供依据。 The fingerprinting of 30 Minnan oolong tea germplasms was constructed based on RAPD molecular marker technology, the results showed the tested tea varieties can be identified by using the same amplimer banding patterns and combination of different amplimer banding patterns; total of 103 bands were generated with 10 RAPD primers, which 80 (77.67%) were polymorphic. Using UPGMA method, the cluster analysis revealed that these tea germplasms were clustered into two groups, and the mean genetic similarity coefficient of tested tea germplasms were 0.603. The genetic relationships of tea germplasms were clearly revealed in the dendrogram, and it would provide a basis for evaluating the genetic evaluation status of the tested samples and choosing appropriate germ- plasm as parents for tea breeding program.
