The expression of telomerase activity in non-small-cell lung cancer and its clinic significance

摘要 目的:探讨端粒酶活性在非小细胞肺癌(NSCLC)中的表达及其作为NSCLC肿瘤标记物和治疗靶点的可能性。方法:采用端粒重复扩增法(TRAP)研究了74例NSCLC组织及周围非癌组织、10例肺良性病变组织端粒酶活性表达。结果:74例NSCLC组织69例(93.24%)有端粒酶活性表达,74例周围非癌组织有9例(12.16%)表达阳性,10例良性病变中2例(20.00%)表达阳性,而相应的病变旁正常组织无端粒酶活性表达,端粒酶活性表达率与肿瘤组织类型、分化程度、肿瘤大小、病理分期无明显相关性。结论:端粒酶活性较普遍存在于NSCLC中,有可能成为NSCLC诊断和判断疗效的标记物和治疗新靶点。 Objective: To investigate the telomerase activity expression in the non-small-cell lung cancer (NSCLC)and the possibility of the telomerase activity as tumor marker and treatment target of NSCLC. Methods: Using PCR-based telomeric repeats amplification protocol(TRAP) assay, we examined telomerase activity in 74 samples of NSCLC and 74 samples of normal lung tissue,10 samples of lung benign lesion and lesion-adjacent tissues. Results:Telomerase activity was detected in 93. 24% (69/74) of NSCLC, 12. 16% (9/74) of normal lung tissues, 20% of the benign lesions(2/ 10 ) and none in benign lesion-adjacent tissues. The frequency of telomerase expression was not associated with histologic types, differentiation, tumor size and pathologic stages. Conclusions: The expression of telomerase is frequent in NSCLC, it may be used as a tumor marker for diagnosis and may become a new target for NSCLC treatment.
