缺氧诱导因子1在糖尿病心肌病大鼠的表达

摘要 目的研究缺氧诱导因子1(HIF-1)在糖尿病心肌病(DC)大鼠心肌的表达,并探讨氯沙坦钾干预对DC大鼠心肌HIF-1表达的影响。方法单次腹腔注射链脲佐菌素(STZ)诱导糖尿病大鼠模型,将糖尿病鼠随机分为DC组(20只)和氯沙坦钾组(L组,20只),以正常大鼠(C组,20只)作对照。干预12周后,观察各组大鼠心率、血糖、脂代谢相关指标、心脏胶原含量变化;用实时荧光定量聚合酶链反应(PCR)分析心肌HIF-1。结果①DC组大鼠心肌HIF-1mRNA水平较正常大鼠明显升高,0.78±0.33 vs 0.33±0.18(P<0.01);而L组大鼠上述指标明显下降,0.54±0.14 vs 0.33±0.18(P<0.05)。②DC组大鼠血糖、血脂均显著高于正常大鼠,干预后上述指标改善不明显。结论氯沙坦钾可能通过下调DC大鼠心肌HIF-1表达,从而发挥其心肌保护作用。 Objective To explore the expression of hypoxia-inducible factovl (HIF-1) in rats with diabetic cardiomyopathy(DC). Methods Diabetic model was established by a single injection of streptozotocin(STZ),diabetic rats were randomly divided into three groups:DC group( n = 20), losartan group(L group, n = 20) and normal control group (NC, n = 20). After intervening for 12 weeks, the heart rate, blood glucose, serum lipid profile and the content of collagen change in the rats were detected, the expressions of HIF-1 were analyzed by real-time Q-PCR. Results Compared with normal group, the HIF-1 mRNA expression in DC group was significantly increased, 0.78 ± 0.33 vs 0. 33±0.18( P 〈0.01),but the HIF-1 expression was significantly decreased in L group 0.54±0.14( P 〈0.05 ). Although blood glucose and serum lipid profile were significantly changed in DC group compared with normal group( P 〈0.05), no significant change occurred following losartan treatment. Conclusion Losartan may down-regulate the expression of HIF-1 in rats with DC and showing the protective effect.
