摘要
目的:优选猫爪草粗多糖提取工艺。方法:采用苯酚-硫酸法测定粗多糖含量(以葡萄糖计),通过正交设计L9(34)优化水提和醇沉工艺。结果:猫爪草粗多糖最佳水提工艺为A3B2C3,即加12倍量水,提取2次,每次3 h;最佳醇沉工艺为A1B1C3,即提取液浓缩至800 g.L-1,加乙醇使含醇量达50%,静置6 h。结论:该方法制备猫爪草粗多糖操作简便、经济、无毒,适用于工业化生产。
Objective:To optimize the best extraction process of crude polysaccharide from Ranunculus ternatus thunb. Methods:Phenol- sulfuric acid method was used to determine the content of crude polysaccharide (based on the content of glucose), orthogonal design L9 (34 ) was used to optimize the best extraction and alcohol precipitation process of polysaccharide. Results : The best extraction process of crude polysaccharide from Ranunculus ternatus thunb was A3B2C3 ,namely extracting twice with adding 12 times water,3 hours every time;the best alcohol precipitation Process was A1B1C3, namely concentrating extraction solution to 0.8 g · mL^-1 , adding ethanol to make the content of ethanol be 50% with standing 6 hours. Conclusion :This method of preparing crude polysaccharide from ranunculus ternatus thunb is simple, economic, non-toxic, and is suitable for industrial mass production.
