摘要
目的通过观察缺血预处理后大鼠局灶脑缺血后神经细胞凋亡变化,探讨脑缺血预处理减少神经细胞凋亡的机制。方法成年健康SD大鼠72只,随机分为预处理组、缺血组、假手术组,用大脑中动脉栓塞法制作脑缺血动物模型,每组在造模成功后24h、48h、72h取出大鼠脑组织,应用原位末端转移酶标记技术(TUNEL染色)观察各个时间观察点细胞的凋亡情况。结果原位末端转移酶标记技术(TUNEL染色)结果显示,缺血组凋亡细胞数最多,预处理组次之,假手术组最少。各组凋亡细胞数随时间呈上升趋势。结论脑缺血预处理可抑制神经细胞凋亡。
【Objective】To explore through observation of ischemic preconditioning in rats after focal cerebral ischemia neuronal apoptosis and cerebral ischemic preconditioning reduces neuronal apoptosis mechanism.【Methods】Method of adult SD rats 72were randomLy divided into the pretreatment group,ischemia group and sham group,middle cerebral artery embolization production of cerebral ischemia animal model,each24 hours after successful modeling,48 h,72 h to remove the rat brain,in situ terminal deoxynucleotidyl transferase labeling(TUNEL staining) observed each time to observe the point of cell apoptosis.The results of the in situ terminal deoxynucleotidyl transferaselabeling(TUNEL staining) 【Resluts】show that the number of apoptotic cells in ischemia group most pretreatment group,followed by sham operation group at least.Upward trendin each group number of apoptotic cells over time.【Conclusion】Cerebral ischemic preconditioning can inhibit neuronal apoptosis.
