树突状细胞/结肠转移癌细胞融合疫苗治疗转移性结肠癌的动物实验研究

摘要 目的通过建立人免疫重建荷人结肠癌皮下种植瘤严重联合免疫缺陷(SCID)小鼠模型,研究DC/结肠癌融合疫苗对人转移性结肠癌的发展及预后的影响。方法应用腹腔注射法对严重联合免疫缺陷SCID小鼠注射人外周血淋巴细胞(hu_PBL)免疫重建后,皮下注射法建立荷人结肠癌皮下种植瘤模型。应用聚乙二醇(PEG)制备DC/SW620融合疫苗。将DC/SW620融合疫苗于左、右后足底、尾根部皮下三点注射在人免疫重建的SCID小鼠结肠癌皮下移植瘤模型。酶联免疫吸附试验(ELISA)检测小鼠血清中人IgG含量;通过直接测量法观察人结肠癌皮下种植瘤成瘤性和小鼠生存期。结果融合疫苗治疗组的皮下肿瘤体积生长速度慢于其他各对照组;融合疫苗治疗组生存期为(109.00±17.17)d,明显长于单纯制模组(55.67±12.03,P<0.001)、免疫重建组(81.83±9.06,P<0.001)、肿瘤细胞疫苗组(87.33±12.85,P<0.01)。结论 DC/SW620融合疫苗可抑制肿瘤生长,改善预后,DC/SW620融合疫苗比肿瘤细胞冻融物免疫治疗效果更佳。
