EEFSEC基因和8q24与中国北京汉族人群前列腺癌患病风险的关系

摘要 目的探索EEFSEC基因(rs10934853,A)和8q24(rs6983561,C;rs445114,T)与中国北京汉族人群前列腺癌(prostate cancer,Pca)临床相关危险因素间的关联性。方法收集212名Pca患者和231名正常对照者血液标本,用病例-对照和单纯病例法进行两组间的等位基因及基因型频率差异分析,探讨各基因SNP与Pca及相关临床表型之间的关联性。结果①EEFSEC基因(rs10934853,A)和8q24(rs6983561,C;rs445114,T)等位基因和基因型频率在病例组和对照组间的分布差异无统计学意义(P>0.05)。②EEFSEC基因(rs10934853,A)和8q24(rs6983561,C;rs445114,T)3个SNPs之间具有协同作用。③基因环境交互作用分析表明携带8q24rs6983561的AA基因型人群Pca易感性与患病年龄负相关(P=0.024),具有统计学意义。其他SNPs与Pca的患病年龄、Gleason评分、PSA浓度以及肿瘤分期等指标均无显著相关性(P>0.05)。结论中国北京汉族人群中EEFSEC基因(rs10934853,A)和8q24(rs6983561,C;rs445114,T)3个SNPs的协同作用可能会增加中国人群的Pca患病风险。携带8q24rs6983561的AA基因型人群Pca易感性与患病年龄负相关。 Objective To explore the associations between 3 SNPs (rs10934853 at EEFSEC and rs6983561, rs445114 at 8q24) and Pca, and the relative clinical risk factors in Chinese Han populations. Methods Blood samples and personal information of 212 Pca patients and 231 controls were collected. The frequencies of alleles and genotypes of these 3 SNPs in two groups were analyzed based on the model of case - control and caseonly studies. We also explored the association between these loci and the clinical characteristics. Results ① There is no significantly different distribution of risk alleles and genotype frequencies in cases and controls among the association of EEFSEC (rs10934853, A) and 8q24 (rs6983561, C; rs445114, T) and Pca(P 〉 0.05).② The interaction of 3 SNPs in EEFSEC (rs10934853, A) and 8q24 (rs6983561, C; rs445114, T) showed that EEFSEC and 8q24 had synergism. ③ The interaction between genes and the environment people with genotype AA on the site of rs6983561 had less probability to get Pca when getting older ( P = 0. 024). Other sites had no significant relevance (P 〈 0.05 ) with the age of Pca on - set, PSA level, Gleason and pathology on indicators. Conclusion Combined EEFSEC (rs10934853, A ) and 8q24 (rs6983561, C; rs445114, T) might raise the risk of Pca of Chinese Han people. People with genotype AA on the site of rs6983561 have less probability to get Pea when getting older.
