摘要
为研究打顶前后烤烟叶片蛋白质表达的差异,利用蛋白质双向电泳分离的方法对烟草打顶前和打顶后10天两个时期的叶片蛋白质表达进行了研究。经过对蛋白质表达图谱的分析和质谱鉴定,分析出9个在两个时期差异表达的蛋白质。在打顶后叶片中高表达的蛋白质有4个,其中3个与光合作用中光反应相关的蛋白,分别是叶绿体放氧复合蛋白,Atfkbp13和PsbP家族蛋白;另外1个打顶后显著表达增加的蛋白是β-1,3-葡聚糖酶前体蛋白,该蛋白与植物发育调控和抗病性有密切关系。在打顶后表达量降低的蛋白共鉴定出5个,其中SHM1,SHM2和乙醇酸氧化酶为与光呼吸相关的蛋白,而叶绿体核糖体蛋白L1和60S核糖体蛋白与植物生长发育和细胞分裂相关。这些差异表达的蛋白表明,打顶可以通过在蛋白水平促进烟草叶片的光合作用和叶片抗性途径相关蛋白的表达,而增强叶片的光合作用和叶片的抗性,同时对与光呼吸能量代谢相关的蛋白表达则有所抑制,从而减弱叶片的能量代谢以调节叶片的生长。
Protein expression levels at 10 days before and after topping were investigated with 2-D electrophoresis. Bioinformatics and MALDI-TOF/TOF analysis showed expression level of nine proteins differ significantly. Four proteins expressed higher after topping, of which the first three were related to photosynthesis, i.e. chloroplast oxygen evolving complex protein, Atfkbpl3 and PsbP family protein. One was prepro-beta-1,3-glucanase precursor which was related to plant growth regulation and plant disease resistance. Five proteins showed lower expression level after topping, of which SHM1, SHM2 and glycolate oxidase were photorespiration-related proteins. Two were ribosomal protein and chloroplast ribosomal protein L1, which were involved in plant growth regulation and cell division. This result indicated that topping can promote photosynthesis and resistance in leaf through increasing expression of photosynthesis and disease resistance related proteins and weaken photorespiration through decreasing expression of energy pathway related proteins.
