摘要
目的建立HPLC-ELSD法同步测定朱砂根活性成分岩白菜素和百两金皂苷A的含量。方法采用Phenomenex Luna C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-水为流动相梯度洗脱,流速1.0 mL min-1,柱温30℃。ELSD参数:漂移管温度100℃,载气流速2.5 mL min-1。结果岩白菜素和百两金皂苷A分别在0.42~8.40μg和0.51~10.20μg与峰面积呈良好的线性关系,平均回收率分别为101.2%、101.4%,RSD分别为1.6%、2.0%。结论建立的含量测定方法操作简便、结果准确、重现性好,可用于朱砂根药材的质量控制。
Objective To establish an HPLC-ELSD method to determine the contents of bergenin and ardisiacrispin A in Ardisia crenata simultaneously.Methods An HPLC-ELSD method was developed.A Phenomenex Luna C18 column(4.6 mm × 250 mm,5 μm) was used,and methanol-water was used as mobile phase with gradient elution,at 1.0 mL ? min-1,The column temperature was 30 ℃.For ELSD,the drift tube temperature was 100 ℃and nitrogen flow rate was 2.5 mL ? min-1.Results Bergenin and ardisiacrispin A were linear at 0.42-8.40 μg and 0.51-10.20 μg respectively.The average recovery was 101.2% and 101.4%,with RSD of 1.6% and 2.0%,respectively.Conclusion The method is easy to operate,accurate and reproducible,which can be used for the quality control of the Ardisia crenata Sims.
