摘要 将5(4,6-二氯三吖嗪)氨基荧光素(DTAF)和普施安红MX-8B活性染料同时标记在淀粉的适当位置,制得了测定淀粉酶活性的荧光新底物。在此底物中,染料将从荧光剂上获得能量转移并使荧光熄灭。淀粉酶催化淀粉的水解,导致能量转移效应降低,荧光强度增大。用此新方法测定了血清中淀粉酶活性,与标准方法比较,相关系数为0.994。 A new method for Serum Amylase Activity assay is developed. The principle of this method is based on the use of a starch sabstrate labeled both with 5-(4, 6-triazin-2-yl)-aminofluorescein (DTAF) and proein red MX-8B dye, which efficiently accept energy from fluorescein, there by quenching fluorescein emission; Amylase Catalyzed Cleavage of labeled starch will increase the average distance between fluorescein and the quencher, leading to a decrease in energy transfer between fluorescein and quencher and an increase in the observed fluorescein emission. The relative fluorescence intensity after incubation time of 5min. or the rate at which fluorescence increases will serve as a measure of Amylase activity. This method shows excellent linearity, very good proportionality between analytical response and amylase activity, good precision in series and superior sensitivity.
