摘要
以不带节的睡菜茎段为外植体,研究其离体再生体系。结果表明:将茎段接种到MS+6-BA 1.0mg/L+IBA 0.5mg/L培养基上10d可以诱导产生愈伤组织,然后转接至MS+6-BA 2.5mg/L+NAA 0.2mg/L+KT 0.05mg/L培养基上15d可使愈伤组织分化为幼苗;将带节的睡菜茎段接种到MS+IBA 0.1~0.2mg/L培养基上7d可使其腋芽萌发为幼苗;将幼苗接种到1/2MS+IBA 0.5mg/L培养基上3~5d可生根,实现植株再生。
Taking the stems without axillary bud of Menyanthes trifbliate explants,its in vitro culture and regeneration were studied. The results showed that putting the stern on the medium MS+6-BA 1.0 mg/L+IBA 0. 5 mg/L for 10 d could produce callus,then put them on MS+6-BA 2. 5 rng/L+NAA 0. 2 mg/L+KT 0. 05 rng/L for 15 days,it could differentiate into seedlings without root;taking the sterns with bud on MS+IBA 0. 1-0. 2 mg/L for 7 days,axillary bud could germinate and then be seeding without root.Put the seedling on 1/2MS+IBA 0. 5 rng/L for 3-5 d,it could rooting.
