摘要
目的对大鼠胰岛进行分离和纯化。方法胶原酶V内消化法分离胰岛,在通过Histopaque分离液1119、1083、1077配合使用HBSS缓冲液对胰岛进行纯化。结果该方法能够得到充足的胰岛,每只大鼠能够获取(758±55)个胰岛,纯度大于95%,并且与Ficoll法相比操作更加简单。结论胶原酶V内消化联合Histopaque法分离和纯化大鼠胰岛是一种简单有效的方法。
Purpose Isolation and purification of rat islets. Method Retrograde perfusion of collagenase V to pancreas in order to isolate islets, and purification of islets were performed using histopaque 1119, 1083, 1077 and HBSS. Result Enough islets were harvested (758 ±55) islets with the purity greater than 95%. This method is relatively easier and more practical compared with the ficoll method. Conclusion Isolation and purification of rat islets by using collagenase V and histopaque is a simple and effective method.
