探针的修饰与浓度杂交温度及时间对基因芯片杂交信号的影响 被引量:3

摘要 本研究以猪链球菌2型的cps2J基因为试验材料,研究了两种探针在不同杂交条件下对基因芯片杂交信号的影响,试验结果表明,寡核苷酸探针的浓度在5~20μm/L之间对杂交信号的影响不大,寡核苷酸探针的氨基化端加上Poly(dT),42℃杂交1h可以获得较好的杂交信号。 In the pagy, our study is focused on the influence of two probes on gene chip hybridization signals under different conditions of hybridization using Streptococcus suis II cps2J gene as an experimental material. Results showed that oligonucleotide probes whose concentration was between 5~20μm/L havelittle effect on hybridization signals;however better hybridization signals can be achieved by adding Poly (dT) to the amino-terminal end of Oligonucleotide probes and hybridizing 1 h in 42 ℃.
