痛风康Ⅱ号颗粒对尿酸钠结晶诱导的急性痛风性关节炎的影响

The Effect of TFKⅡ Granule on MSU-induced Acute Gouty Arthritis

目的观察痛风康II号颗粒对尿酸钠结晶(MSU)致急性痛风性关节炎大鼠关节红肿、功能障碍和滑膜组织炎症病理改变的影响。方法 48只大鼠随机分为正常组、模型组、秋水仙碱组、痛风康Ⅱ号颗粒高剂量组、痛风康Ⅱ号颗粒中剂量组、痛风康Ⅱ号颗粒低剂量组,每组8只,正常组和模型组采用生理盐水灌胃,疗程5 d。用药第3天,除正常组外所有大鼠均采用尿酸钠溶液关节腔注射法造模,正常组注入相同体积的生理盐水。动态测量大鼠右踝关节周径,观察大鼠炎症指数及功能障碍指数,HE染色法观察大鼠关节滑膜组织的病理变化。结果与正常组相比,模型组造模后4,10,24,72h关节肿胀指数均显著增加(均P<0.01)。与模型组相比,痛风康Ⅱ号颗粒高、中、低剂量组造模后4,10,24 h关节肿胀有减轻的趋势,72 h关节肿胀均显著减轻(均P<0.01),痛风康Ⅱ号颗粒高、中剂量组关节肿胀指数有较低剂量组降低的趋势。模型组10 h表现出明显的关节炎症红肿和功能障碍,到24,72 h表现出逐步缓解的趋势;与模型组比较,痛风康Ⅱ号颗粒低、中、高剂量组关节炎症红肿和功能障碍明显减轻。HE染色显示模型大鼠关节滑膜炎症明显,滑膜细胞增生,伴炎细胞浸润,小血管增生及成纤维细胞,经痛风康Ⅱ号颗粒治疗后,上述病理改变减轻。结论痛风康Ⅱ号颗粒可明显改善MSU致急性痛风性关节炎模型大鼠的关节肿胀、红肿及功能障碍,改善滑膜组织炎症病理变化。

Objective To research the effect of TFK I granule on joint swelling,joint inflammation,joint dysfunction and pathological changes of synovial tissue inflammation in rats with MSU - induced acute gouty arthritis. Methods 48 rats were randomly divided into normal group,model group,colehieine group,large dose TFK Ⅱgranule group, medium dose TFK Ⅱ granule group, small dose TFK Ⅱ granule group ,8 rats in each group. Saline water was administered to the normal group and model group. All rats had been orally administered by the drugs for 5 days. On the third day of administration, acute gouty arthritis model was set up via articular cavity MSU injection except the normal group, the same volume of saline was injected to the normal group. Right ankle joint circumference of rats was measured dynamically. The indexes of rats＇inflammation and dysfunction were observed. The pathological changes of rats joint synovial tissue were detected by HE staining. Results Compared with normal group, the joint swelling index of model group was significantly increased （ 4h, 10b, 24h, 72h after model ： all P 〈 0.01 ）. Compared with model group, the joint swelling index of TFK H granule large, medium and small dose groups showed a decreasing trend at 4h, 10h and 24h, but was significantly lightened（ all P 〈0.01 ） ,and the joint swelling index of TFK Ⅱ granule large and medium dose groups showed a decreasing trend compared with small dose group. Model group showed significant joint inflammation and joint dysfunction （10h） , and a gradually catabatic trend（24h,72h） ;Compared with model groups,the joint inflammation and joint dysfunction of TFK ]] granule small, medium and large dose groups was significantly lightened. The abnormalities of the ratg joint synovial tissue included obvi- ous inflammation, synovial cells proliferation, inflammatory cell infiltration, small vessels proliferation, fibroblast cell by HE staining, which showed the animal model was successfully produced. After the treatment with TFK Ⅱ granule, these histological abnormalities were lessened. Conclusion TFK Ⅱ granule can significantly improve joint swelling,inflammation, dysfunction of rats with MSU -induced acute gouty arthritis, and can improve pathological changes in synovial tissue inflammation.
