摘要
目的对枸杞中类胡萝卜素类成分进行了提取方法、提取溶剂及正交实验等系统性研究,并测定枸杞中类胡萝卜素清除DPPH的能力。方法以β-胡萝卜素为对照品,采用正交实验方法研究了溶剂比例、提取时间、提取次数、固液比等对枸杞中类胡萝卜素提取得率的影响。以清除DPPH自由基为指标测定了枸杞中类胡萝卜素的清除自由基能力。结果最佳提取工艺为:水浴加热回流50℃提取3次,每次1 h,提取溶剂为石油醚∶丙酮(7∶3)。枸杞中类胡萝卜素对DPPH的清除率IC50值为37.2μg/ml。结论优化了枸杞中类胡萝卜素的提取工艺,枸杞中的类胡萝卜素具有较好的抗氧化能力。
Objective To optimize the extraction and to determine the antioxidant activity for carotenoid in Lycium barbarum L. Methods With β - carotene as a reference, orthogonal experiment was designed from the reflux with temperature, heating time, extracting times and the ratio of gardenia to liquor. Results Carotenoid concentration in Lycium barbarum L. was extracted the highest when the water bath calefaction reflux extracting three times, heat time 3 hours, petroleum etheracetone (7:3), heating temperature 50℃. Conclusion The inhibitory rate of DPPH scavenging activity (IC50 = 37.2μg/ml) was analyzed for carotenoid in Lycium barbarum L.
