摘要
目的观察中药中霉菌毒素对NIH/3T3细胞中DNA聚合酶β(DNApolβ)表达量的影响,研究中药不良反应的机制和互隔交链孢酚致细胞变异的机制。方法体外培养NIH/3T3细胞,用不同浓度的霉菌毒素——互隔交链孢酚(AOH)加入培养液影响细胞。用RT-PCR方法从细胞全RNA中扩增出DNA聚合酶β基因cDNA序列,同时扩增β-actin进行半定量分析了解其表达量的变化。结果成功从NIH/3T3细胞中扩增了DNA聚合酶β基因cDNA,通过半定量分析发现随着药物浓度的增高,DNA聚合酶β表达量上调。结论霉菌毒素可导致细胞中的DNA聚合酶β表达量上调从而影响基因修复系统的功能,AOH的作用浓度越高,DNA聚合酶β基因表达量越高。
Objective To observe the expression of DNA polymeraseβ(DNApolβ) in NIH/3T3 cell line affected by alternariol (AOH), and study on the mechanism of ahernariol inducing cancer. Methods Affect the cells by AOH with different concentration. DNApolβ gene was amplified through RT -PCR from the cells, and amplified β -actin at the same time,then detect expression account of DNApolβ gene. Results The expression account was increased with the higher concentration of AOH. Conclusion The higher concentration of AOH acts,the more change of DNApolβ gene expression can be seen.
