摘要
毛状根培养是由发根农杆菌转化植物建立起来的体系,具有激素自养、生长迅速、化学成分含量与原植物相当或高于原植物及生物合成能力相对稳定等优点,近年来已在多种植物中建立,并开始用于重组药物蛋白的合成,不但为植物化学成分的大规模生产开辟了新的途径,而且使那些在细菌、酵母菌和哺乳动物体内难以表达的药物蛋白的合成成为了可能。文章系统综述了近年来毛状根培养在植物化学成分和重组药物蛋白研究方面的进展和成果,以期为毛状根培养的深入研究与开发提供指导。
Hairy root cultures (HRCs) ,induced by Agrobacterium rhizogenes infection and cultured individually on phytohormone - free medium, have been established from numerous plant species in recent years. HRCs grow lastly, accumulate phytochemicals whose levels are comparable or higher than that of intact plants and are usually stable in their biosynthetic capacities. Hairy roots can be grown in industrial - scale bioreaetors providing abundant and sustainable sources of phytoehemieals. HRCs have also been used in the synthesis of recombinant therapeutic proteins, especially those hard to express in bacteria, yeast and mammalian expression systems. This review presents recent findings in phytoehemieal and recombinant protein production by the utilization of HRCs so as to guide its further research and development.
