依托咪酯或异丙酚麻醉对大鼠海马神经元特异性核蛋白表达的影响

摘要 目的:探讨依托咪酯或异丙酚麻醉对大鼠海马神经元特异性核蛋白(NeuN)表达的影响。方法:随机将雄性SD大鼠140只分为对照组、异丙酚Ⅰ组、异丙酚Ⅱ组、异丙酚Ⅲ组、依托咪酯Ⅰ组、依托咪酯Ⅱ组、依托咪酯Ⅲ组,每组20只。异丙酚Ⅰ组、异丙酚Ⅱ组、异丙酚Ⅲ组腹腔注射异丙酚总量分别为50,100,200mg/kg,依托咪酯Ⅰ组、依托咪酯Ⅱ组、依托咪酯Ⅲ组腹腔注射依托咪酯总量分别为10,30,60mg/kg,对照组不注射任何药物。血气分析仪检测大鼠动脉血呼吸和代谢指标的变化,免疫组化法检测大鼠海马CA1区、CA3区和齿状回NeuN的表达。结果:各组大鼠动脉血pH、氧分压(PaO2)、二氧化碳分压(PaCO2)、HCO3-、BE、氧饱和度(SaO2)之间比较差异无统计学意义(P>0.05);与对照组比较,异丙酚Ⅰ组、依托咪酯Ⅰ组、异丙酚Ⅱ组、依托咪酯Ⅱ组大鼠海马CA1区NeuN阳性神经元平均光密度值差异无统计学意义(P>0.05),而异丙酚Ⅲ组、依托咪酯Ⅲ组大鼠海马CA1区NeuN阳性神经元平均光密度值减少(P<0.05),异丙酚Ⅲ组和依托咪酯Ⅲ组比较差异无统计学意义(P>0.05);各组大鼠海马CA3区和齿状回的NeuN阳性神经元平均光密度值比较差异无统计学意义(P>0.05)。结论:较高剂量依托咪酯和异丙酚可降低大鼠海马CA1区神经元NeuN的表达,但对CA3区和齿状回无影响。 Objective. To observe the effects of etomidate or propofol on the expression of NeuN in hippocampal neurons of rats. Methods. A total of 140 male Sprague Dawley rats aged 4 w were randomly divided into 7 groups ( n =20). The rats in propofol I , II and III groups were intraperitoneally injected with 50, 100 and 200 mg/kg propofol, respectively; and the rats in etomidate I , II and III groups were intraperitoneally injected with 10, 30 and 60 mg/kg etomidate, respectively; and the rats in control group were injected with no drugs. Breathing and metabolic indexes were detected by blood gas analyzer. The expression of NeuN in CA1, CA3 and dentate gyrus regions of hippocampus was observed by immunohistochemical method. Results: Breathing and metabolic indexes were not different among seven groups. However, 60 mg/kg of etomidate or 200 mg/kg of propofol decreased NeuN-positive neurons in hippocampal CA1 region. Etomidate or propofol didn't affect the expression of NeuN in CA3 and dentate gyrus regions of hippocampus. Conclusion: High dose of etomidate or propofol may decrease the expresion of NeuN in hippocampal CA1 region of rats.
