仙虎合剂预防大鼠实验性脂肪肝的实验研究

Study on xianhu mistura in preventing experimental fatty liver in rats

目的观察仙虎合剂对大鼠实验性脂肪肝的预防作用。方法将wistar大白鼠50只随机分为正常对照组、高脂对照组、仙虎合剂高、低剂量组和甲硫氨酸组。从实验第1天起,除正常对照组外,其余四组均以高脂饮食喂养,并都同时给予相应剂量的药物灌胃。正常对照组、高脂对照组用白开水灌胃,仙虎合剂高、低剂量组用仙虎合剂(分别相当于生药31.5 g.kg-1,15.75 g.kg-1)灌胃,甲硫氨酸组用甲硫氨酸片(0.625 g.kg-1)灌胃。连续用药10周后处死取材送检,观察血清TC、TG、LDL-C、肝组织内脂质及形态学变化等。结果与高脂对照组相比,仙虎合剂高、低剂量组血清及肝组织内TC、TG值明显降低,血清LDL-C显著降低(P<0.01或P<0.05),肝细胞的脂肪变性程度及细胞肿胀坏死程度明显减轻,且作用与甲硫氨酸相当。结论仙虎合剂可以调节血脂,对高脂饮食诱导的实验性脂肪肝的形成具有明显的预防作用。

Objective To observe the preventive effect of xianhu mistura on experimental fatty liver in rats.Methods 50 Wistar rats were divided into normal control group,high fat control group,high dose xianhu mistura group,low dose xianhu mistura group,methionine group.From the first day of experiment,in addition to the normal group,the other four groups were fed high-fat diet,given the corresponding medicine by gavage.The normal control group and high fat control group were given plain boiled water by gavage,the high dose xianhu mistura group were given xianhu mistura（31.5 g·kg-1） by gavage,the low dose xianhu mistura group were given xianhu mistura（15.75 g·kg-1） by gavage,the methionine group were given methionine tablets（0.625 g·kg-1） by gavage.After 10 weeks of continuous medication,all rats were examined to observe the effect of xianhu mistura on serum TC、TG、LDL-C、the lipid in liver and morphological changes of experimental fatty liver in rats.Results Compared with the high fat control group,xianhu mistura could significantly lower serum TC、TG、LDL-C（P0.05 or 0.01）,lighten the degree of hepatic and hepatocell necrosis of wistar rats.And quite with the methionine group.Conclusion Xianhu mistura could regulate blood lipid,and has better effect on the prevention of hyperlipidemia fatty liver in rats.
