摘要
目的:比较三个药对及全方地格达-4味汤对急性肝损伤大鼠的生物效应,进而验证地格达-4味汤配伍的合理性和科学性.方法:取150只大鼠,按体重随机分正常对照组、模型组、阳性对照组、肋柱花加栀子大中小三个剂量组、肋柱花加胡黄连大中小三个剂量组、肋柱花加瞿麦大中小三个剂量组、地格达-4味汤大中小三个剂量组等15组.给药14天后,各给药组大鼠腹腔注射D-半乳糖胺(D-GlaN)300mg/kg进行造模,采用全自动生化分析仪测定各组血清天门冬氨酸氨基转移酶(AST)和丙氨酸氨基转移梅(ALT)含量,继而比较药对及全方血清AST和ALT含量.结果:地格达-4味汤可明显降低D-GlaN致肝损伤大鼠的血清AST和ALT含量(P<0.05).肋柱花加栀子药对大和中剂量组AST含量明显降低,差异显著(P<0.05);而ALT含量无明显变化(P>0.05).肋柱花加栀子小剂量组未能明显降低血清AST、ALT含量(P>0.05).药对肋柱花加瞿麦和肋柱花加胡黄连组均未能明显降低血清AST、ALT含量(P>0.05).结论:地格达-4味汤全方对D-GlaN致急性肝损伤的保护作用优于药对.因此,本实验可提示地格达-4味汤配伍的合理性和科学性.
Objective: Through comparing the hepatoprotective efficacy of Digda-4 decoction with its couplet medi- cines on rat acute liver injury model, to verify the reasonableness and seientificalness of Digda-4 decoction' s compati- bility. Methods: 50 rats were randomly divided into 15 groups according to their weight, including normal control, model, positive control, combination of Lomatogonium rotatum with Gardenia (in high, moderate and low doses), com- bination of Lomatogonium rotatum with Picrorhiza' (in high, moderate and low doses), combination of Lomatogonium ro- tatum with Fringed pink (in high, moderate and low doses), and Digda-4 decoc4ion (composed of all 4 herbs shown above; in high, moderate and loy~ doses). After 14 days of administration, D-galactosamine (D-GlaN) at 300mg/kg was intraperitoneally injected into all rats (except normal control group) to establish acute liver injury model. The aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in the rat serum among all groups were detected 24 hours later by automatic biochemical analyzer and their levels were compared. Results: The serum concentrations of AST and ALT in D-GlaN induced rat liver injury reduced significantly in Digda-4 decoction group (P〈0.05). The se- rum level of AST in the combination group of Lomatogonium rotatum with Gardenia at high and middle doses also re- duced significantly (P〈0.05), while ALT did not change significantly (P〉0.05). This combination at low dose did not influence AST and ALT levels at all (P〉O.05). The serum AST and ALT in the combination group of Lomatogonium ra- tatum with Fringed pink, and the combination group of Lomatogonium rotatum with Picrorhiza did not reduced signifi- cantly either (P〉0.05). Conclusion: Digda-4 decoction is superior to its couplet medicines in hepatoprotective effects on D-GIaN induced acute rat live injury. The experimental result implies the reasonable and scientific compatibility of Digda-4 decoction.
