全抗原MQCA-BSA耦联比HPLC测定方法的建立

摘要 为了建立一种准确、可靠的测定人工抗原耦联比的方法,试验应用高效液相色谱(HPLC)法采用Waters 2695分离系统、2489紫外检测系统,流动相为甲醇、水之比为60∶40,柱温为30℃,流速为1.00 mL/min,检测波长为320 nm,测定全抗原合成中游离3-甲基喹啉-2-羧酸(MQCA)的含量。结果表明:游离MQCA的线性范围为0.2~128.0μg/mL,线性方程为A=17 100 C+16 667(R2=0.999 5)。说明本方法准确、可靠,适用于人工抗原偶联比的测定。 To establish an accurate and reliable method for determination of the coupling ratio of artificial antigen, the content of free 3 - meth- ylquinoline evil -2 - carboxylic acid (MQCA) in the synthesis of the complete antigen was determined by Waters 2695 and 2489 UV HPLC system. In the experiment, the mobile phase was methanol, the water ratio was 60:40, the column temperature was 30 ℃, the flow rate of 1.00 mL/min, and the detection wavelength was set at 320 rim. The results showed that the linear range for the free MQCA were from 0.2 to 128.0 g/mL, and the linear equation was A = 17 100C + 16 667 (R^2 = 0. 999 5 ). The results indicate that the method is accurate, reliable, and suitable for the determination of the coupling ratio of the artificial antigen.
