GC法同时测定维药芜菁子中4种异硫氰酸酯类化合物的含量

Content Determination of 4 Kinds of Isothiocyanates Compounds in the Seeds of Brassica rapa by GC Me- thod

目的:建立同时测定维药芜菁子中异硫氰酸酯类化合物含量的方法。方法:采用水蒸气蒸馏法提取芜菁子中的异硫氰酸酯,以异辛醇为内标物,利用气相色谱法分析异硫氰酸酯类化合物的组成并测定其含量。结果:芜菁子中含有4种异硫氰酸酯类化合物,分别为异硫氰酸苯乙酯(14.20%)、异硫氰酸叔丁酯(0.47%)、异硫氰酸异丙酯(0.29%)和异硫氰酸烯丙酯(5.10%)。4种化合物的进样量线性范围依次为10.06～50.30μg(r=0.9996)、0.25～1.26μg(r=0.9995)、2.83～14.15μg(r=0.9989)、3.34～16.72μg(r=0.9986);平均加样回收率分别为99.2%、100.2%、97.8%、98.2%,RSD分别为1.3%、1.9%、2.3%、2.1%(n均为9)。结论:本方法简便、灵敏、专属性强、重复性好,可用于芜菁子的质量控制。

OBJECTIVE： To establish the method for the content determination of isothiocyanate compounds in the seeds of Brassica rapa. METHODS： Isothiocyanates was extracted from the seeds of B. rapa by steam distillation method. Using isooctanol as internal standard, its components and contents were analyzed by GC. RESULTS： There were 4 kinds of isothiocyanates in the seeds of Brassica rapa, such as phenylhexyl isothiocyanat （14.20%）,text-butyisothiocyanate （0.47%）, isopropyl isothiocyanate （0.29%）,allyl isothiocyanate（5.10%）. The linear ranges of 4 compounds were 10.06～50.30 μg（r=0.999 6）,0.25～1.26 μg（r=0.999 5）,2.83～14.15 μg（r=0.998 9） and 3.34～16.72 μg（r=0.998 6）, respectively. Average recovery rates were 99.2%, 100.2%, 97.8% and 98.2%, and RSDs were 1.3%, 1.9%, 2.3% and 2.1%（n=9）, respectively. CONCLUSION： The method is simple, sensitive, specific and reproducible. It can be used for quality control of the seeds of B. rapa.
