基于气相色谱-质谱联用技术的转人血清白蛋白水稻的代谢组学方法研究

摘要 为推动代谢组学研究,利用转人血清白蛋白水稻,对样品前处理、色谱柱选择、GC-MS的检测方法、谱图处理和数据分析等方面进行研究。结果显示:在提取液为水∶乙腈∶异丙醇=1∶1.5∶1.5的条件下,30℃衍生90min,作为样品前处理条件,选用DB-5MS色谱柱,采用保留时间锁定的数据采集方法,用AMDIS软件的保留时间分析模式对谱图解卷积,然后经过MPP软件进行主成分分析,建立了一套完整的代谢组学研究方法流程。 In order to promote the metabolomic research,the transgenic rice with human serum albumin was studied by using the sample pretreatment,chromatographic column choice,gas chromatography-mass spectrometry(GC-MS)detection method,spectrogram analysis,and data analysis.The results showed that the pretreatment conditions of the samples were the extract conditions of water∶acetonitrile∶isopropanol = 2∶3∶3,and was derivative in 90 minutes at 30℃.DB-5 MS chromatographic column and the data acquisition method of the retention time locked were adopted,while AMDIS software of retention time analysis type was used to find components in the labrary.And then after MPP software to do principal component analysis,a complete process of metabolomic research methods was finished.
