摘要
为分离和鉴定出猪圆环病毒2型(PCV2)贵州株,对送检的3份感染病料进行了分离,并将其分离毒株分别同步接种于PK-15细胞,再特异性扩增和克隆所分离毒株的全基因序列。结果表明:成功分离出3株PCV2贵州株,命名为GZ-QZ1株(1 767bp)、GZ-RH1株(1 767bp)和GZ-CS1株(1 768bp);3株PCV2贵州株全基因组之间的核苷酸相似性为94.6%~99.9%,其中ORF2核苷酸相似性为90.2%~99.9%;ORF2氨基酸一致性GZ-QZ1和GZ-RH1为100%,二者与GZ-CS1的氨基酸一致性均为88.1%。3株PCV2贵州株之间同源,但GZ-CS1存在一定变异。
In order to obtain PCV2 Guizhou strains,three samples,which were detected PCV2 positive,were inoculated PK-15 cells by using synchronous inoculation method,and the whole gene of the isolated PCV2 strains were specific amplified and cloned,then sequenced.The results showed that the three PCV2 Guizhou strains,which were named GZ-QZ1 strain(1 767 bp),GZ-RH1 strain(1 767 bp) and GZ-CS1 strain(1 768 bp),were successfully isolated and their complete genome were identified.The nucleotide similarity of the complete genome of the above three strains were 94.6%~99.9%,the nucleotide similarity of ORF2 gene were 90.2%~99.9%.The consistency of amino acids encoded by ORF2 gene was 100% between GZ-QZ1 strain and GZ-RH1 strain,and the consistency was 88.1% between GZ-CS1 strain and these two strains,this result indicated that these three strains are homologous,but some variation existed in GZ-CS1 strain.
