摘要 【目的】探讨Pim-1抑制剂对巨噬细胞pNF-kBp65、iNOS,TNF-a表达的影响。【方法】在脂多糖(LPS)刺激前给予不同浓度的Pim-1特异性抑制剂PIM-Inh,WesternBlot检测pNF-kBp65的表达,RT—PCR测定iNOS,TNF—d的表达,比较不同浓度抑制剂对pNF-kBp65、iNOS,TNF-a表达的影响。【结果】用不同浓度PIM-Inh预处理后,NF-kBp65、iNOS,TNF-a的表达明显下降(P〈0.05),且呈剂量依赖性,浓度100μmol/L时最为明显。【结论】PIMmInh特异性抑制剂可能抑制TLR4/NF-kB信号转导,降低TNF-a,iNOS的表达。 [Objective]To explore the effect of Pim-1 kinase inhibitor(PIM-Inh) on the expression of pNF- kB p65, iNOS and TNF-a in macrophage. [Methods] Before lipopolysaccharide(LPS) stimulation, different concentrations of PIM-Inh were given. The expression of pNF-kB p65 was detected by Western blot. RT-PCR was used to detect the expression of iNOS and TNF-a. The effects of different concentrations of PIM-Inh on the expression of pNF-kB p65, iNOS and TNF-a were compared. [Results] After the pretreatment with PIM- Inh 10μmol/L and 100/μmol/L, the expression of pNF-kB p65, iNOS and TNFa obviously decreased in a dose- dependent manner( P d0.05). The effect of PIM-Inh 100/μmol/L was most obvious. [Conclusion]PIM-Inh may inhibit TLR4/NF-kB signal transduction and reduce the expression of TNF-a and iNOS.
