摘要
从生活污水取样,以大豆油为惟一碳源,通过富集驯化培养、中性红油脂平板初筛得到29株菌株.经透明圈法筛选,得到7株透明圈直径(D)与菌落直径(d)比值大于或等于2的菌株.测定各菌株发酵液的脂肪酶活性进行复筛,得到一株高效产脂肪酶菌株X2,其脂肪酶活性为41.7 U/mL,所产脂肪酶稳定性较好.根据其形态学及生理生化特征,初步鉴定该菌株为产碱假单胞菌.
Using bean oil as the sole carbon source, 29 lipase - producing strains were isolated from wastewater through enrichment and domesticating method. Seven strains out of the 29 isolates were selected which showed the D/d ratio≥2 by the larger transparent zone. Strains was cultured in fermentation culture medium in which the mass concentration of bean oil was 20g/L. Among the 7 strains. Strain X2 showed the highest lipase activity of 41.7 U/mL and better lipase stability. Based on morphological, physiological and biochemical characteristics, strain X2 was identified as Pseudomonas alcaligenes.
