胰腺癌患者术前与术后血清蛋白质组学研究 被引量:1

Serum proteomics analysis on patients with pancreatic cancer before and after operation

摘要 目的利用比较蛋白质组学方法分离、鉴定胰腺癌患者术前、术后血清差异表达蛋白质,筛选鉴定出特异性候选标志物。方法采用双向差异凝胶电泳(2D-DIGE)技术分离2007年5月至2008年9月期间安徽医科大学附属省立医院收治的20例胰腺癌患者外周血清蛋白质,利用基质辅助激光解析电离飞行时间质谱(MALDI—TOF—MS)技术对差异蛋白质进行鉴定。结果成功鉴定出5个胰腺癌差异表达蛋白质:结合珠蛋白(HP)、载脂蛋白E(ApoE)、补体C481、TNIP2蛋白和血清淀粉样蛋白P成分A链。该组蛋白质在胰腺癌术前组呈高表达,在胰腺癌术后组呈低表达。各点术前表达分别较术后高4.16、2.28、1.62、1.78、1.55倍(P〈0.05)。结论应用蛋白质组学技术筛选胰腺癌患者术前、术后外周血清中的特异性生物标志物的方法快速、有效。 Objective To separate and identify protein markers for serum differentially expressed proteins in patients with pancreatic cancer before and after operation by using proteomic method. Methods Differentially expressed proteins were separated from peripheral blood in 20 patients with pancreatic cancer patients before and after operation from May 2007 to September 2008 by 2D-DIGE, and identified by using SELDI-TOF-MS. Results Five differentially expressed proteins: haptoglobin protein (HP protein), apoli- poprotein E (ApoE), CAB1, isoform 1 of TNFAIP3-interacting protein 2 (TNIP2) and chain A (the struc- ture of pentameric human serum amyloid P component) were identified by 2D-DIGE and SELDI-TOF-MS. Those proteins had higher expression before operation than after operation in the patients with pancreatic cancer, and preoperative expression at each point was 4. 16, 2. 28, 1.62, 1.78, and 1.55 times higher than that after operation, respectively ( P 〈 0. 05 ). Conclusion Proteomics technology is a quick, easy and practical method to screen for specific biomarkers in serum of patients with pancreatic cancer.
