摘要
目的:研究去窦弓神经(Sinoaortic denervated,SAD)对自发性高血压大鼠(Spontaneous hypertensive rat,SHR)主动脉瞬时受体电位通道C3(Transient receptor potential canonical 3,TRPC3)及TRPC6亚型表达的影响,探讨TRPC3及TRPC6与血压变异(Blood pressure variability,BPV)的关系。方法:14只10周龄雄性SHR随机分为SAD组和假手术组,各7只。术后8周在清醒、自由活动状态下测定两组大鼠24 h动脉血压及BPV,采用逆转录多聚酶链反应(Reverse transcription polymerase chain reaction,RT-PCR)法检测主动脉组织TRPC3及TRPC6信使核糖核苷酸(Messenger ribonucleic acid,mRNA)的表达,蛋白免疫印迹法(Western blot)与免疫组织化学法(Immunohistochemistry,IHC)检测其蛋白的表达。结果:术后8周,两组大鼠24 h平均收缩压(Systolic blood pressure,SBP)和舒张压(Diastolic blood pressure,DBP)比较无显著性差异;SAD组24 h收缩压变异(Systolic blood pressure variability,SBPV)显著高于假手术组大鼠(P<0.05),SAD组24 h舒张压变异(Diastolic blood pressure variability,DBPV)亦显著高于假手术组大鼠(P<0.01),主动脉组织TRPC3及TRPC6的mRNA和蛋白表达均显著高于假手术组大鼠(P<0.01),差异均有统计学意义。结论:SAD组大鼠SBPV和DBPV升高,同时TRPC3及TRPC6的mRNA和蛋白表达升高,提示TRPC3及TRPC6的升高与其BPV升高密切相关,TRPC3及TRPC6可能参与BPV的调控。
Objective:To investigate the effects of sinoaortic denervated(SAD)on the expressions of transient receptor potential canonical 3(TRPC3)channel and TRPC6 channel in the aorta of spontaneous hypertensive rats(SHR)and to discuss the relationship among TRPC3,TRPC6 and blood pressure variability(BPV).Methods:Fourteen male SHR at the age of 10 weeks were randomly divided into two groups:SAD operation group(n=7)and sham operation group(n=7).24 h blood pressure and BPV were measured in conscious,unrestrained rats at 8 weeks after operation.Expressions of messenger ribonucleic acid(mRNA)and protein of TRPC3 and TRPC6 were determined by RT-PCR,Western blot and immunohistochemistry staining respectively.Results:There was no difference in 24 h average systolic blood pressure(SBP)and diastolic blood pressure(DBP)between the two groups;24 h systolic blood pressure variability(SBPV)and diastolic blood pressure variability(DBPV)were significantly higher in SAD group than in sham operation group(P0.05),(P0.01);expressions of mRNA and protein of TRPC3 and TRPC6 were significantly higher in SAD group than in sham operation group(P0.01).Conclusions:SBPV and DBPV as well as mRNA and protein expressions of TRPC3 and TRPC6 of rats in SAD group are increased,indicating that the increased expressions of TRPC3 and TRPC6 are closely associated with elevated BPV and might be involved in the regulation of BPV.
