非小细胞肺癌组织中p38蛋白的表达及其与吸烟的关系 被引量:1

摘要 目的:检测非小细胞肺癌组织中p38的表达,探讨其与患者吸烟的关系。方法:应用固定化蛋白质印迹法检测52例非小细胞肺癌及癌旁正常肺组织中p38的表达情况;免疫组织化学法分析其在细胞内的定位。结果:肺癌组织中p38的表达水平与癌旁正常组织无明显差别(P>0.05);27例(51.9%)标本肺癌组织中p38蛋白的表达水平高于癌旁正常组织(P>0.05);p38的表达在腺癌组织中明显增加,与肺癌肿瘤直径大小、淋巴结转移及TNM分期无关。而不吸烟患者癌组织标本中p38的表达水平较吸烟者高,为吸烟者的2.3倍(P<0.01);不吸烟者腺癌组织中p38的表达水平显著增高。免疫组织化学显示p38分布在胞浆内。结论:p38在肺腺癌不吸烟者中过表达,可能在不吸烟者的肺癌发生过程中起着重要作用。 Objective:To investigate the expression of mitogen-activated protein kinase p38 and its relationship with smoking in non-small cell lung cancer.Methods: Samples were obteined from 52 patients with non-small cell lung cancer.Western blot was used to measure the expression of p38.Immunohistochemistry was used for localization of p38.Results: There was no difference of the expression of p38 in lung cancer compared with those in adjacent normal tissues(P0.05).In 27 samples,the expression level of p38 in lung cancer tissues was increased(P 0.05).The expression level of p38 was found no correlation with tumor size,TNM staging,lymph node involvement,but it was correlated with pathologic type,and overexpressed in adenocarcinoma.The expression level of p38 increased in no smoking patients,and it was 2.3 times compared with those of smoking patients.Further study showed that in no smoking patients,the expression level of p38 was higher than those of squamous cell carcinoma.Immunohistochemistry showed that p38 was located only in cytoplasm.Conclusion: p38 overexpressed in adenocarcinoma patients without smoking,and it maybe play an important role in the development of human non-small cell lung cancer patients never smoking.
