荧光定量PCR检测非小细胞肺癌中MTA1和E-cadherin的表达及意义 被引量:3

Significance of the expression of MTA1 and E-cadherin genes in non-small cell lung carcinoma

摘要 目的探讨肿瘤转移相关蛋白家族1(MTA1)和E-钙黏蛋白(E-cadherin)在人非小细胞肺癌(NSCLC)中的表达及意义。方法采用实时荧光定量PCR(Q-PCR)联合检测40例NSCLC组织、12例癌旁组织、12例良性病变肺组织中MTA1和E-cadherin mRNA表达的相对水平,分析他们的表达与临床组织病理学特征的关系。结果MTA1 mRNA在NSCLC中平均表达水平高于癌旁和良性疾病组织(P<0.05);E-cadherin mRNA在NSCLC中平均表达水平低于癌旁和良性疾病组织(P<0.05);在NSCLC中MTA1和E-cadherin的表达呈负相关(r=-0.561,P<0.05);在癌旁组织和良性疾病组织中MTA1和E-cadherin的表达无统计学差异(P>0.05);MTA1和E-cadherinmRNA表达水平与淋巴结转移及临床分期有关(P<0.05)。结论 MTA1过度表达及E-cadherin表达的下调或缺失与NSCLC浸润和转移呈正相关,联合检测NSCLC中MTA1和E-cadherin的表达,可用于预后评估。 Objective To explore the expression of MTA1 and E-cadherin genes and their significance in human non-small cell lung cancer (NSCLC). Methods Employ the real-time fluorescence quantitative polymerasc chain reaction ( Q- PCR) to examine the expression of MTA1 and E-cadherin mRNA in 40 samples of NSCLC tissues, 12 samples of paracarcinoma tissues and 12 samples of benign lung tissues, and to evaluate the relativity of MTA1, E-cadherin expression and NSCLC clinical pathologic features. Results The average expression level of MTAI mRNA in NSCLC was higher than that in the paracarcinoma tissues and the benign lesions tissue ( P 〈 0.05 ). The average expression level of E-cadherin in NSCLC was lower than that in the paracarcinoma tissue and the benign lesions tissue ( P 〈 0.05 ). In NSCLC tissues the ex- pression of MTA1 and E-cadherin was negatively correlated (r = -0. 561, P 〈 0.05 ). There was no statistical difference in the MTA1 and E-cadherin expression between the cancer and benign disease tissue. The expression of MTAI and E-cadherin mRNA was closely relevant to lymph node metastasis and the clinical stages ( P 〈 0.05 ). Conclusions The overexpression of the MTA1 gene and the loss or lack expression of the E-cadherin were correlated with invasion and metastasis of NSCLC. Both of the expression of MTA1 and E-cadherin could work together for prognosis assessment.
