超高效液相色谱-串联质谱法研究荭草提取物中4个黄酮类成分的大鼠组织分布 被引量:5

摘要 目的:建立同时测定大鼠静脉注射荭草提取物后组织器官中荭草素、牡荆素、木犀草苷和槲皮苷含量的UPLC-MS/MS方法,研究荭草提取物在大鼠体内心、肝、脾、肺、肾等9个组织器官的分布特征。方法:色谱柱为BEH C18柱(2.1 mm×50mm,1.7μm),流动相:0.1%甲酸乙腈-0.1%甲酸水,梯度洗脱,流速0.35 mL.min-1。组织样品机械匀浆后甲醇沉淀供测定。结果:在线性范围内4个成分的线性关系良好,准确度、日内和日间精密度、提取回收率均符合生物样品的分析要求。大鼠静脉注射荭草提取物后的组织分布试验结果表明:4个成分在肾脏等9种组织器官中均能检测到。4个成分主要分布在肾、肺等组织中,在脑中分布最少。结论:本文方法可用于荭草花提取物在大鼠体内的组织分布研究。 Objective: To establish an UPLC -MS/MS method for simultaneous determination of orientin, vitexin, cynaroside and quercitrin in different tissues of rats after a single administration of Polygonum orientale L. extract by intravenous injection,and to investigate the distribution of the four compounds in 9 organs of rats such as heart,liv- er, spleen,lung, kidney and others. Methods: The UPLC gradient elution was performed using BEH Cl8 (2. 1 mm×50mm,1.7μm)column with the mobile phase of 0. 1% acetonitrile -0. 1% formic acid at the flow rate of 0. 35 mL · min^-1. The tissue samples were homogenated and the suspension was extracted by precipitating methanol for determination. Results:Tile relationships between the concentration and the peak areas of the four compounds were all linear. The precisions, accuracy, extract recoveries and stability of the analytes met the biological requirements. It was demonstrated that the four compounds could be detected in all concerned tissues and a high concentration of the four components were mainly obtained in kidney and lung, while the lowest concentration was found in brain after the single intravenous administration. Conclusion : The method can be used to the study of tissue distribution of Po- lygonum orientale L. extract in rats.
