摘要
目的:建立麦冬药材的TLC及HPLC特征图谱,为科学评价和有效控制其质量提供可靠方法。方法:采用3种TLC法建立麦冬薄层特征图谱;采用HPLC法建立麦冬液相特征图谱:色谱柱为Welch AQ C18色谱柱(200 mm×4.6 mm,5μm),乙腈-水为流动相,梯度洗脱,柱温30℃,流速为1.0 mL.min-1,检测波长205 nm,进样量10μL。结果:麦冬TLC鉴别分离良好;麦冬HPLC特征图谱有6个共有峰,不同来源的川麦冬相似度大于0.94,3批浙麦冬相似度低,HPLC方法的精密度、稳定性和重现性良好。结论:所建方法可行,可用于麦冬药材的质量控制。
Objective: To establish the TLC and HPLC characteristic chromatograms of Ophiopogonis Radix, so as to provide a reliable method for scientific evaluation and effective quality control of this medicinal material. Methods: Three kinds of TLC method were adopted to establish the thin - layer characteristic ehromatogram of Ophiopogonis Radix. And the HPLC method was used to establish the liquid Characteristic chromatogram of Ophiopogonis Radix. The HPLC analysis was performed on a Welch AQ C18column (200 mm ×4.6mm,5μm). The mobile phase was acetonitrile - water with gradient elution. The flow rate was 1.0 mL · min ^- 1 and the detection wavelength was 205 nm, the sample volume 10 μL, and the column temperature was 30 ℃. Results: Ophiopogonis Radix was well identi- fied and separated. The HPLC characteristic ehromatogram of Ophiopogonis Radix had 6 common peaks. Sichuan Ophiopogonis Radix from different places of origins were of high similarity ( 〉 0. 94), while the Ophiopogonis Radix of 3 batches were of low similarity. The HPLC method was precise, stable and reproducible. Conclusion : These es- tablished methods are practical and suitable for the quality control of Ophiopogonis Radix.
