摘要
目的建立以HPLC-ELSD对黄芪提取物中单糖和二糖进行测定的方法。方法采用PrevailTMCarbohydrate ES色谱柱,乙腈-水(75∶25)为流动相,柱温35℃,流速0.8 mL.mim-1;ELSD漂移管温度60℃,载气为空气,气压25 psi。结果黄芪提取物中的单糖和二糖组分能够得到较好地分离,果糖和蔗糖的线性范围分别为0.052~1.042、2.442~48.84 mg,r分别为0.9947、0.9965;平均加样回收率分别为98.6%、102.9%,RSD分别为1.2%、1.0%。结论所用方法快速简捷、结果准确稳定,适合于黄芪提取物中单糖、双糖的含量测定。
OBJECTIVE To establish an HPLC-ELSD method for the simultaneous quantitative analysis of fructose and sucrose from Astragalus extracts.METHODS The chromatographic separation was achieved on PrevailTM Carbohydrate ES column using a mobile phase composed of a mixture of acetonitrile and water(75:25) at a flow rate of 0.8 mL·mim-1.The ELSD drift tube temperature was set at 60 ℃ and the carrying gas was air whose pressure was 25 psi.RESULTS Monosaccharides and disaccharides from Astragalus extracts could be well separated,and the linear ranges of fructose and sucrose were 0.052-1.042 mg and 2.442-48.84 mg,and the average spiked recoveries of fructose and sucrose were 98.6%(RSD=1.2%) and 102.9%(RSD=1.0%).CONCLUSION This method is accurate,rapid,simple and reproducible,and it is suitable for the determination of monosaccharides and disaccharides in Astragalus extracts.
