摘要
目的探讨Cyclin D1和hMSH2在结直肠癌组织中的表达及其与结直肠癌临床病理特征之间的关系。方法用免疫组化SP法检测50例结直肠癌组织中Cyclin D1和hMSH2的表达,用Kaplan-Meier法分析Cyclin D1和hMSH2的表达与临床病理特征和预后的相关性。用多因素COX模型分析影响患者预后的相关因素。结果在正常组织、癌旁组织和癌组织中Cyclin D1蛋白表达阳性率分别为0、40%和64%,而hMSH2蛋白表达阳性率分别为10%、52%和86%。在结直肠癌组织中Cyclin D1和hMSH2蛋白表达率明显高于正常组织,差异有统计学意义(P<0.01)。Cyclin D1和hMSH2在结直肠癌中表达与分期和分化程度及2年内复发率有关,差异有统计学意义(P<0.05),而与病理类型无关。多因素分析显示,Cyclin D1和hMSH2(RR值分别为2.405和2.312,P<0.05)的表达是结直肠癌的重要预后因素。结论 Cyclin D1和hMSH2基因突变可能与结直肠癌的发生、发展和预后有一定关系。
Objectlve To study the expression of Cyclin Dland hMSH2 in colorectal cancer and their relationship with the elinieopathologieal features of the disease. Methods The expression of Cyelin D1 and hMSH2 were observed in 50 cases of eoloreetal cancer tissues using immunohistochemical staining. Kaplan-Meier survival analysis was used to analyze the correlation of the expression of Cyclin D1 with the clinieopathologic parameters and prognosis of colorectal cancer. Multiple COX proportional hazards model analysis was used to analyze the prognostic factors. Results The positive rates of Cyelin D1 protein expression in normal tissue, paraeaneer organizations and cancer organizations were 0, 40% and 64%, respectively, and the positive rates of hMSH2 protein expression were 10%, 52% and 86%. The positive expression of them were related with cellular differentiation, TNM stage and the recurrence rate in two years, but it was not related with the pathological type. Multiple analyses showed that Cyclin D1 and hMSH2 expression was important prognostic factors for coloreetal cancer. Conclusion Cyelin D1 and hMSH2 gene mutations may he associated with the occurrence, development and prognosis of colorectal cancer.
