“消瘀散”外用剂型的体外透皮特性比较研究

摘要 目的:通过对"消瘀散"几种剂型体外透皮试验研究,比较不同剂型的经皮渗透特性,为药理、毒理试验及临床研究提供依据。方法:以有效成分大黄素累积透皮百分率为测定指标,高效液相色谱法定量,用改良的Franz扩散池及离体兔皮进行体外透皮实验,比较几种剂型透皮速度、透皮百分率与皮肤储量的差别。结果:透皮速度由快至慢依次为:即型凝胶≈乳膏>贴膏剂>散剂;透皮总量由多至少依次为:乳膏>即型凝胶>贴膏剂>散剂;皮肤储量由高到低依次为:即型凝胶>乳膏>贴膏剂>散剂>涂膜剂。结论:即型凝胶透皮速度最快,皮肤储量最多,乳膏透皮总量最多。 Objective : Through the research of transdermal permeation in vitro of several kinds of dosage forms of Xi aoyu Powder, comparison among transdermal permeation characteristics in different dosage forms was made, providing the base for pharmacology, toxicology experiment and clinical research. Method: The accumulated penetration rate of the emo-din( a kind of effective component)was used as an indicator. The quantity was determined by HPLC. The improved Franz diffusion cells and rabbit skin in vitro were used as the method that can take the test of penetration, comparing the differ- ences among the penetration rate, the percentage of transdermal permeation and the reserves in skin of several kinds of dosage forms. Result : The rate of penetration transform from fast to slow : followed by in - situ gel ≈ emulsifiable paste 〉 emplastrum 〉 powder ; the total amount of penetration transform from fast to slow : followed by emulsifiable paste 〉 in - situ gel 〉 emplastrum 〉 powder ; the reserves in skin transform from fast to slow : followed by In - situ gel 〉 emulsifiable paste 〉 emplastrum 〉 powder. Conclusion : The fastest penetration rate and the most reserve in skin is in - situ gel, the most total amount is emulsifiable paste.
