摘要
目的:观察蝎毒素(ITX)对Lewis肺癌生长及对荷瘤小鼠免疫细胞的影响。方法:建立Lewis肺癌荷瘤小鼠模型后,随机分为模型组、rIL-2组、ITX组、空白组,每组8只小鼠,接种第2日起,每天腹腔注射给药,连用16天。末次给药24h后,用流式细胞术检测小鼠外周血中免疫细胞的功能状态;酶联免疫法检测小鼠外周血TGF-β1水平,并测量瘤体质量和体积。结果:ITX干预后荷瘤小鼠单核、NK和B细胞均有一定活化,但无统计学意义;rIL-2干预后荷瘤小鼠的NK和B细胞有明显活化(P<0.05);ITX组和rIL-2组血清TGF-β1水平均高于模型组,rIL-2组明显高于模型组(P<0.05);ITX和rIL-2干预后瘤体质量和体积均有增加,ITX组明显高于模型组(P<0.05)。结论:ITX和rIL-2均促进了瘤体质量和体积的增加,可能与其活化的免疫细胞有关。
Objective:To investigate the effect of scorpion venom (ITX) on tumor growth and imnmne cells of Lewis Lung Carcinoma in C57BL/6 Mice. Methods : Holland Lewis lung carcinoma mice were taken as the model and randomly divided into four groups:the model group, the rlL-2 group, the ITX group, the normal control group. There were 8 mice in each group. Each group was inoculated intraperitoneally since the second day of inoculation for 16 days. Twenty-four hours after the last injection, the level of immune cells in peripheral blood was analyzed by flow cytometry ;the contentence of TGF-β1 in peripheral blood serum was de- tected by enzyme-linked immune sorbent assay ; subcutaneous tumors were measured for tumor weight and volume. Results : The activation of monocyte, NK and B cells were promoted by ITX, without statistics difference.( P 〉 0. 05). The levels of NK and B ceils were elevated by rIL-2 (P 〈0. 05 ). The level of TGF-β1 was elevated by ITX and rIL-2 ,but only rIL-2 group was statistical different( P 〈 0. 05 ). ITX and rlL-2 promoted tumor weight and volume, but only ITX group was statistical different (P 〈 0. 05 ). Conclusion : ITX and rlL-2 can promote tumor growth. It may be related to the activation of immune cells.
