摘要
通过比较各种分析柱的分离效果,建立了用离子交换柱分离,邻苯二甲醛(OPA)柱 后衍生荧光检测分析生物组织中组胺的液相色谱方法,并对流动相的离子强度和pH进行了 优化,确定以40mmol/L pH5.50的柠檬酸钠缓冲液作为流动相为最佳条件。该方法的检测限 为50 nmol/L,回收率在 92%以上,线性范围为 50 nmol/L~ 500 μmol/L。分析时间缩短为 15 min。
A rapid and simple method for the determination of histamine in biological sample by postcolumn derivatization liquid chromatography with fluorescence detection was described. A SCX weakly acidic cation exchange column was used as an analytical column. Histamine was eluted at 13. 2 min by 40 mmol/L of trisodium citrate (pH 5 .50) at flow rate of 1. 0 mL/min. The recoveries of histamine from 1 .0 μmol/L to 100 were above 92%. The detection limit was 50nmol/L and the linear of the determination was 50 nmol/L ~ 500 μmol/L histamine.
