摘要
目的探讨可卡因-苯丙胺调节转录肽(Cocain and am-phetamine regulated transcript peptides,CART)对脑缺血小鼠脑组织8-羟基-2’-脱氧鸟苷(8-hydroxy-2’-deoxyguanosine,8-OHdG)和3-硝基酪氨酸(3-nitrtyrosine,3-NT)水平的影响。方法健康雄性昆明白小鼠随机分为4组:脑缺血组(每时间点n=15),CART组(每时间点n=15),生理盐水(NS)对照组(每时间点n=15),假手术组(每时间点n=12)。建立大脑中动脉闭塞(MCAO)模型,CART组、生理盐水对照组于缺血2h即刻分别经尾静脉注射给予CART55-102(0.5μg)或同体积NS。在脑缺血不同时间点分别进行脑梗死体积和脑组织8-OHdG及3-NT水平检测。结果脑缺血后不同时间点脑组织8-OHdG及3-NT含量均被上调,CART可下调缺血后脑内8-OHdG及3-NT水平的升高,缩小梗死体积。结论 CART能通过下调8-OHdG及3-NT水平,抑制氧化应激,有效保护小鼠缺血性脑损伤。
Objective To investigate the effects of CART on level of 8-OHdG and 3-NT after cerebral ischemia injury in mice.Method Healthy male Kunming white mice were randomly divided into four groups:cerebral ischemia group(every point n=15),CART group(every point n=15),normal saline(NS)control group(every point n=15)and sham-operation group(every point n=12).A middle cerebral artery occlusion(MCAO)model was induced.Two hours after MCAO,CART55-102 injection and equivalent NS were injected via tail vein in CART group and control group respectively.Infarct volume and the expression of 8-OHdG and 3-NT were detected at different time points after MCAO.Results The content of 8-OHdG and 3-NT were upregulated at different points after cerebral ischemia in mice.CART could significantly downregulate the increase of 8-OHdG and 3-NT in brain,and reduce infarct volume.Conclusion CART may effectively protect against ischemic brain injury in mice,and its mechanism is associated with inhibiting the expression of 8-OHdG and 3-NT,which are the productions of protein and nucleotide peroxidation.
