摘要
以常年石油浸渍土壤为筛选原料,通过驯化、筛选和诱变的方法选育出对甲胺磷农药降解最显著的菌株。通过在培养基中先后加入不同浓度的甲胺磷农药进行菌株的梯度驯化,以驯化后菌株对甲胺磷农药的降解率为指标,筛选出降解效果最好的菌株XY-C,经紫外照射诱变后测得菌株对甲胺磷农药的降解率为79.2%,且六次传代稳定性良好。通过对菌株的形态学观察,生理生化实验,透射电镜鉴定,确定经紫外诱变后得到的XY-C-3为葡萄球菌属(Staphylococcus rosehbach)。
Used the soil as raw material ,which was impregnated in petroleum perennially. The most remarkable methamidophos pesticides degradated strains were selected through the domestication,screening and mutagenesis. Different concentrations methamidophos pesticides were added into the culture medium early or late for the gradient domestication of the strain. Took the ratio of the domesticated strain degradated methamidophos pesticides,the most effect strain were selected as XY-C. After ultraviolet radiation irradiated, the degrated ratio of methamidophos pesticides was 79.2%,the heredity stability was better after six times. The XY-C-3 strain,which was after UV mutagenesis,was identified as Staphylococcus rosehbach by morphology observation, physiologybiochemistry test and transmission electron mocroscopic images.
