摘要
为了得到花生中引起过敏反应的主要致敏成分Ara h1,以新鲜花生为材料,通过粉碎、脱脂、硫酸铵分步盐析等方法进行粗提;并用离子交换柱以及凝胶柱等方法来进一步纯化过敏原Ara h1。采用聚丙烯酰胺凝胶电泳(SDS-PAGE)分析了纯化后的过敏原Ara h1的纯度,并用高效液相色谱法测定其纯度达到90%以上。
In order to get the main sensitization composition Ara hl in peanuts,which can cause allergic reactions. Fresh peanuts were chosen as the material and some methods including crushing,skimming,ammonium sulfate by salting-out,dialysis were adopted to purify the main allergen Ara hl. And at last it was purified by DEAE ion-exchange column and SuperdexTM 200 gel column. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and high performance liquid chromatography(HPLC) method were used to determine the purified allergen Ara hl ,and the results showed that the purity was above 90%.
